Why a high peak percentage need not tell you the amount of material present.

Different questions

Chromatographic purity asks how much of the detected signal is attributed to one component under a stated method. Content asks how much of a specified substance is present in a sample or container. Water, counterions and other constituents can affect mass-based quantities without appearing as equivalent peaks in a peptide chromatogram.

Value assignment needs more

A study of synthetic human C-peptide compared methods for assigning purity mass fraction and identifying related impurities. It shows the need to state the measurand and method before comparing numbers. A label's milligram claim should be supported by a suitable quantitative test on the relevant batch and format.

Research takeaway

Do not convert an HPLC area percentage into milligrams of peptide without an appropriate quantitative method.
