Why activity at several receptors needs separate measurements.
Several targets, several assays
A peptide designed to engage GLP-1, GIP and glucagon receptors presents more than one pharmacological question. A cellular study compared mono-, dual- and tri-peptidic agonists using engineered cells expressing specified receptors and measured signalling pathways. The results depended on those experimental systems.
Balance is not one number
An activity value at one receptor does not describe the other two. To understand a claimed multi-receptor profile, ask for the identity of the test ligand, the receptor panel, reference compounds and comparable assay conditions. Cell measurements alone do not predict a human outcome.
Look for a receptor-by-receptor data set rather than a single “triple agonist” label.
