Cell-culture medium can change a peptide experiment

The material added to a well may not remain unchanged throughout the assay.

Medium is part of the system

Serum, enzymes and other medium constituents can affect peptide persistence. A study using methionine enkephalin compared degradation and activity in cell-culture systems with different serum conditions. It showed that medium composition mattered to the interpretation of that particular experiment.

Check the observation window

If an assay measures a response hours after addition, the concentration present at the end may differ from the starting concentration. Appropriate controls can separate a biological effect from material loss or degradation. Report medium composition, incubation time and the method used to check stability when those variables are relevant.

Research takeaway

An in-vitro result describes a defined medium and time window; it should not be detached from those conditions.
